Calu-3 cells show a combined secretory and ciliated phenotype. Although the Calu-3 cell line comes from a submucosal adenocarcinoma of the bronchi, it is considered a suitable in vitro model of the upper airway epithelium. This cell line is considered a sensitive and efficient preclinical model to study human respiratory processes and diseases ( 22). Upon stimulation with viruses or environmental toxins, the Calu-3 cell line synthesizes and releases different cytokines, including IL-6 ( 20), which play a central role in the inflammatory cascade associated with more severe COVID-19 ( 21). Calu-3 is a cell line derived from a submucosal adenocarcinoma of the bronchi that grows in adherent culture and displays epithelial morphology. Various studies have proposed the need to study SARS-CoV-2 infection in human respiratory epithelium to get closer to the central target tissue of the disease in patients ( 19). The Vero E6 cell line (African green monkey kidney) is deficient for type 1 interferons (IFNs) and highly susceptible to many different pathogens, like measles virus, rubella virus, arboviruses, adenoviruses, influenza, and some coronavirus, including SARS-CoV-2 ( 16– 18). It has recently been described that iota-carrageenan has activity against the SARS-CoV-2 virus and its Spike Pseudotyped Lentivirus (SSPL) in Vero E6 cell culture ( 14, 15). Based on these observations, the hypothesis has been raised that a nasal spray with iota-carrageenan could be effective against SARS-CoV-2. Iota-carrageenan formulated into a nasal spray has already been proven safe and effective in common cold treatment ( 13). It has also been shown that their inhibitory activity also relies on affecting the viral replication cycle at different steps, like entry and genome replication, and additionally activates the host's antiviral immune response ( 10– 12). Iota-carrageenan inhibits virus infection mainly based on its interaction with the surface of viral particles, preventing them from entering cells and trapping the viral particles released from the infected cells ( 9). Besides these properties, the iota-carrageenan demonstrated antiviral activity against several viruses, including respiratory viruses such as human rhinovirus, influenza A H1N1, and common cold coronavirus ( 6– 8). Marine-derived polysaccharides, such as carrageenans, are a family of linear sulfated polysaccharides extracted from red seaweeds, widely used as thickening agents and stabilizers for food. Repurposing established medications with recognized safety profiles is a possible approach for preventing or treating the disease and shortening the time-consuming drug development stages while the vaccination is being extended.ĭuring the first days of the infection, the virus replicates mainly in the nasal cavity and the nasopharynx therefore, nasal sprays with antiviral activity could prevent infection or, in case it occurs, could reduce the viral load in these zones ( 4, 5). Vaccines are the only tool that has been shown to be effective in preventing COVID-19 at the time of this publication, <40% of the world population has received a complete vaccination, and in developing countries, the percentage of individuals with complete vaccination is negligible, and only 3.1% of the population has received a single dose ( 3). There are, except for remdesivir, no approved antivirals for the treatment or prevention of SARS-CoV-2 infection. The severe acute respiratory coronavirus 2 (SARS-CoV-2) is responsible for the currently ongoing pandemic coronavirus disease (COVID-19), counting more than 243,857,028 confirmed cases and more than 4.953.246 deaths worldwide by Octo( 1, 2). In this study, the assayed formula, already approved as a nasal spray for human use, effectively inhibited SARS-CoV-2 infection, providing a more substantial reference for clinical studies or developments. In Calu-3, a human respiratory epithelium cell line, we explored the activity of a formula of iota-carrageenan and sodium chloride against SARS-CoV-2. Iota-carrageenan is a sulfated polysaccharide extracted from red seaweeds, which, formulated into a nasal spray, has already been proven safe and effective in viral upper respiratory infections. While the new restaurant has a vastly different menu than the Fosters Freeze that once occupied the location, the owners decided to pay tribute to the old establishment by installing an ice cream window off the side of the entrance. Even the oil that the food is cooked in is made from organic sunflower oil, she said. Lesh estimates that customers are enjoying meals that are made from 95% organic ingredients. By partnering with numerous local farmers to provide ingredients for the restaurant’s menu, Ms. Lesh and her husband are offering a menu of healthy meals in the heart of Carpinteria. The renovation effort continued through this year and was completed through a crowdsourcing effort on Kickstarter, where community members raised over $50,000 for the construction of the restaurant’s outdoor patio and inventory. Just days before the taping, Unplugged producer Alex Coletti visited the band during a stop on the In Utero tour.Īlex Coletti (producer, MTV Unplugged ): We flew into somewhere in Massachusetts, went to a hotel, and dropped our stuff off, and drove for a bit. We didn’t quite know how many songs they had that would work and how well they would translate to the acoustic setting. So when they were invited to do the show, we had some big questions to figure out. And I had experience with it, having done one with R.E.M.ĭave Grohl (drums) : We’d seen the other Unpluggeds and didn’t like many of them, because most bands would treat them like rock shows-play their hits like it was Madison Square Garden, except with acoustic guitars.Įarnie Bailey (guitar tech): It became a running joke. During that time, all the rage was MTV’s Unplugged show. Scott Litt (producer, MTV Unplugged in New York ): The only thing I thought might be tricky was trying to play Nevermind songs in that acoustic vein. His creative mind at that time was going more in a quieter direction. Mark Kates (A&R, Geffen Records): Kurt wanted to prove to himself that he could do this in an artistically successful way. Everybody was just really excited when they agreed to do it. The story of Nirvana Unplugged began 25 years ago, when MTV’s influence on popular culture was at its peak.Īmy Finnerty (vice president of music and talent, MTV): Everybody wanted to see Nirvana do an Unplugged. You’re lucky if you get to be at something like that once in your lifetime.” “You knew for sure that history was being made,” said former MTV executive Amy Finnerty, who worked closely with Nirvana. As the show progressed, those in attendance began to realize that what they were watching would become legendary. The room’s haunting vibe later led the event to be described as sorrowful, but despite Cobain’s well-documented struggles at the time, the evening was far from dour. The lead singer even helped design the set, asking for it to be decorated with stargazer lilies and black candles. He also invited two of his musical heroes, Cris and Curt Kirkwood of the little-known Meat Puppets. Nirvana’s appearance on the acoustic series proved something that close observers already knew: The loudest band on earth had a stunning amount of depth.Ĭobain subtly subverted the format, which usually featured acts playing stripped-down versions of their hits, by filling the set list with cover songs. Wearing a fuzzy cardigan, ratty button-down, Frightwig T-shirt, jeans, and Converse sneakers, Kurt Cobain-with help from drummer Dave Grohl, bassist Krist Novoselic, guitarist Pat Smear, and cellist Lori Goldston-orchestrated a performance that was heartfelt, funny, uncomfortable, and mesmerizing. That night, the biggest group of the decade staged one of the most hypnotically intimate rock concerts ever captured on film. The other children don't let Sunny (a goody-two-shoes) enter and she is worried they might be doing something dangerous, so Lucy decides to go take a look. She also sees her friend Sunny, who tells her that some of their friends led by Red are secretly playing in Barn Haystack. Out of her house ( Alley), Lucy collects the Tights left hanging to dry, which combine with the Broken Slingshot to make a Slingshot. In First Floor, Eloise asks Lucy to do some shopping around the town and collect the 4 ingredients marked in the List to prepare food for the festival. It's the day of the Harvest Festival, the most important festival in the town.Īlley: In her Bedroom, she collects a Broken Slingshot. Going to bed Lucy hears the growls of the monsters coming from the window. Home: After dinner, Lucy is shown enjoying learning how to sing from her mother, collecting a note. The sun goes down, and while looking for her cat Lucy is frightened by some shadows (one of the monsters?) and sprints back home. Lucy also breaks the Handle from a door and uses it as a clapper to repair the door's bell, collecting a note. Interacting with the trombone will also let Lucy collect the Trombone sticker. Town: In Alley Lucy meets an old crazy lady ( Angela), who tells her that all other adults are cowards (foreshadowing later reveals), and gives her a sticker ( Trinket). She fails and drops, at which point Eloise suggests she go outside to play a bit (father is against it). Lucy then goes in the Cellar to collect some Sausages (which requires to grab and light the Candle in the fireplace first) and coming back she eavesdrops from her parents.Īs a final task, she is requested to collect salt from on top of the cabinet (close to the entrance door), and needs the Stool to reach it. In Second Floor interacting with the mirror Lucy can get the Mirror sticker. (There are 4 songs in the game, each made of 3 notes = first note of the first song = first song etc…) In her Bedroom Lucy finds also her Diary (all notes and stickers go here), while using the Toy Mouse on Squinty in First Floor lets her gain her first note. Lucy is asked to bring a Saucepan for cooking (close to the entrance door) and then Firewood for the fireplace, which requires her to collect the Toy Mouse from her Bedroom upstairs. Home: At Home Lucy finds her father ( Cole) and the cat ( Squinty). Her mother ( Eloise) comes to pick her up soon after and they go home. Historically, the concept of a proof and its associated mathematical rigour first appeared in Greek mathematics, most notably in Euclid's Elements. The problem of integer factorization, for example, which goes back to Euclid in 300 BC, had no practical application before its use in the RSA cryptosystem, now widely used for the security of computer networks. Other areas are developed independently from any application (and are therefore called pure mathematics), but often later find practical applications. Some areas of mathematics, such as statistics and game theory, are developed in close correlation with their applications and are often grouped under applied mathematics. Although mathematics is extensively used for modeling phenomena, the fundamental truths of mathematics are independent from any scientific experimentation. Mathematics is essential in the natural sciences, engineering, medicine, finance, computer science and the social sciences. These results include previously proved theorems, axioms, and-in case of abstraction from nature-some basic properties that are considered true starting points of the theory under consideration. A proof consists of a succession of applications of deductive rules to already established results. These objects consist of either abstractions from nature or-in modern mathematics-entities that are stipulated to have certain properties, called axioms. Most mathematical activity involves the discovery of properties of abstract objects and the use of pure reason to prove them. There is no general consensus among mathematicians about a common definition for their academic discipline. These topics are represented in modern mathematics with the major subdisciplines of number theory, algebra, geometry, and analysis, respectively. Piecewise functions, limits, and continuity are typically studied in advanced math courses, such as calculus or analysis. Continuity is important for understanding how a function behaves and for making predictions about its values. A function is considered continuous if it can be drawn without lifting the pen from the paper.
